Summary:

The mission of the BWH Medicinal Chemistry Core is to serve the Partners HealthCare and non-Partners research communities providing expertise and knowhow to help investigators engage Medicinal Chemistry in their basic or translational studies in an efficient and cost-effective manner.

  • Analytical Services ( Material analysis service )

    Quality control of delivered compounds obtained from commercial vendors or other sources to establish their chemical purity and structural integrity.
    1H-, 13C-, 19F-NMR spectra, as well as 2D-NMR spectra
    • Mass Spectroscopy: high resolution and low resolution MS
    • Purity level by analytical HPLC

  • Consulting Services ( Support service )

    • Selection of libraries for screening
    • Selection of druggable hits
    • Literature and database searches
    • Search for commercially available derivatives or fragments for preliminary biological evaluation
    • Design of structure-activity relationship (SAR) studies
    • Design of synthetic pathways
    • Hit-to-lead and lead optimization
    • Outsourcing custom synthesis when needed
    - Selection of providers
    - Solicitation and analysis of quotes
    - Assessment of synthetic pathways feasibility and cost-efficiency
    - Design and development of statement-of-work (SOW) and deliverables specifications

    • Preliminary testing of alternative synthetic pathways for feasibility and cost-efficiency

    • Custom synthesis of focused libraries and derivatizations for SAR studies
    • Improvement of physicochemical properties of lead candidates and molecular probes
    • Scaling-up synthesis of molecular probes and lead candidates to support in vitro and preliminary in vivo studies
    • Purification and analytical characterization of small molecules by MS and NMR
